Sometimes, calyptra cells (= root cap cells) cannot easily be distinguished form tannin cells. Certainty can be obtained, when the root cap cells are observed at the very tip of the root. They are mostly filled homogeneously and often slightly yellowish. Their shape changes can be followed to the sides of the mycorrhiza.
